Physical Questions Posed by DNA Condensation

چکیده
When multivalent salts are added to dilute DNA solutions, the DNA chains condense into bundles of a well-defined size. We explore two physical problems motivated by this phenomenon: the origin of the attractive interactions between the highly negativelycharged DNA chains that cause them to concentrate into bundles, and the mechanism that prevents the bundles from growing indefinitely large.

Effect of chromatin condensation on frozen-thawed sperm DNA integrity in normozoospermic men
Background and Aim: Sperm cryopreservation is a common technique used for management of male infertility. But this method has detrimental effects on sperm DNA and chromatin quality. Evaluation of different markers associated with the health of genetic material of sperm is beneficial for determination of the fertility of sperm. The aim of this study was to assess the effect of sperm chromatin co...
متن کاملRole of tension and twist in single-molecule DNA condensation.
Using magnetic tweezers, we study in real time the condensation of single DNA molecules under tension. We find that DNA condensation occurs via discrete nucleated events. By measuring the influence of an imposed twist, we show that condensation is initiated by the formation of a plectonemic supercoil. This demonstrates a strong interplay between the condensation transition and externally impose...
متن کاملAbrupt transition from a free, repulsive to a condensed, attractive DNA phase, induced by multivalent polyamine cations.
We have investigated the energetics of DNA condensation by multivalent polyamine cations. Solution small angle x-ray scattering was used to monitor interactions between short 25 base pair dsDNA strands in the free supernatant DNA phase that coexists with the condensed DNA phase. Interestingly, when tetravalent spermine is used, significant inter-DNA repulsion is observed in the free phase, in c...
متن کاملRestriction Enzyme Analysis of DNA Methylation in “Condensed” Chromatin of Ha-Ras-Transformed NIH 3T3 Cells
Increased amounts of chromatin condensation (i.e., localized areas of high DNA density, or chromatin higher order packing state) have been described in NIH 3T3 cells transformed with the Ha-ras oncogene. The structural basis for this oncogene-mediated alteration in nuclear organization is unknown. Since DNA methylation is likely to be involved in regulating the nucleosomal level of DNA packagin...
متن کاملCondensation of DNA-actin polyelectrolyte mixtures driven by ions of different valences.
Multivalent ions can induce condensation of like-charged polyelectrolytes into compact states, a process that requires different ion valences for different polyelectrolyte species. In this work we examine the condensation behavior in binary anionic polyelectrolyte mixtures consisting of DNA coils and F-actin rods in the presence of monovalent, divalent, and trivalent ions. As expected, monovale...
